Immunogenicity of Bacillus Calmette-Guérin in pigs: potential as a translational model of non-specific effects of BCG

چکیده
Background Clinical and immunological studies in humans show that the live attenuated Bacillus Calmette-Guérin (BCG) vaccine has beneficial non-specific effects, increasing resistance against diseases other than tuberculosis. The underlying mechanisms are currently being explored. pig exhibits considerable physiological similarity to anatomy physiology, suggesting similar responses BCG could be expected. Studies of effects pigs scarce. We investigated feasibility using as a large animal model investigate BCG. Methods In series experiments, we randomized newborn or young piglets from conventional farms receiving placebo persistence bacteria various tissues, immunogenicity ex vivo blood vitro stimulation assays, acute phase protein clinical heterologous infectious challenge with influenza A virus Actinobacillus pleuropneumoniae . Results was generally well tolerated. contrast humans, no skin reaction form abscesses, ulcers, scars observed. Live recovered draining lymph nodes 2/13 animals 20 weeks after vaccination. Specific IFN-γ antigen-specific re-stimulation mycobacterial antigen were increased but not TNF-responses TLR2 TLR4 agonists. few genes differentially expressed vaccination, including antiviral RIG-I CSF1 , although effect disappeared correction for multiple testing. symptoms bacterial viral respiratory infections did differ, nor copies nasopharyngeal samples challenge. However, response significantly reduced BCG-vaccinated A. Discussion safe pigs, inducing specific responses, our corroborate innate responsiveness seen humans. dose challenges may have been sub-optimal. Even so, infection animals.

Variability of Bcg Strains (bacillus Calmette-guérin)
Three different cultures of BCG propagated for over 2 years in a liquid medium containing Tween 80 and albumin were found to differ in several of their intrinsic properties. Cultures of the three strains were found to consist of morphologically heterogeneous populations-each culture being made up of three main colonial types-spreading, intermediate, and non-spreading. The percentage distributio...
متن کاملDisseminated Bacillus Calmette-Guérin Disease after BCG Vaccination: A Case Report from China
Background: Bacillus Calmette-Guérin (BCG) vaccination is recommended for newborn infants worldwide to prevent tuberculosis. However, complications do occur inevitably in a very low rate, among which the most serious is disseminated disease. The disseminated bacillus Calmette–Guérin disease is a rare disease with high fatality, and can be seen among persons with an underlying immunodeficiency. ...
متن کاملBacillus Calmette-Guérin (BCG) Infection Following Intravesical BCG Administration as Adjunctive Therapy For Bladder Cancer
Bacillus Calmette-Gu erin (BCG) is the most effective intravesical immunotherapy for superficial bladder cancer. Although generally well tolerated, BCG-related infectious complications may occur following instillation. Much of the current knowledge about this complication comes from single case reports, with heterogeneous diagnostic and therapeutic approaches and no investigation on risk factor...

since the 1960s the age effects on learning both first and second language have been explored by many linguists and applied linguists (e.g lennerberg, 1967; schachter, 1996; long, 1990) and the existence of critical period for language acquisition was found to be a common ground of all these studies. in spite of some common findings, some issues about the impacts of age on acquiring a second or...

Addition of normal C57BL/6 mouse bone marrow cells to an in vitro culture of normal C57BL/6 spleen cells and allogeneic P815-Y tumor cells inhibited the development of cell-mediated immunity. Bacillus Calmette-Guérin (BCG) enhanced the suppressive activity of these bone marrow cells as early as 2 days after its intravenous administration to donor mice and elicited similar activity in the spleen...
